Surface Plasmon Resonance

Surface plasmon resonance is a surface sensitive spectroscopic technique that measures changes in the refractive index of a medium in direct contact with a nanoparticle surface. It involves propagating electron density waves that occur at the interface between a metal and dielectric. When a polarized light beam is transmitted through a prism with a deposited nanoparticle film and reaches a specific angle greater than the angle of total internal reflection, a sharp minimum is observed in the reflection coefficient due to losses in the metal, creating surface plasmons. SPR can be used as a sensitive technique for pH, temperature, pressure, and other environmental sensors as well as for protein detection. It offers advantages such as being label-free, reusable, and allowing real-time monitoring.

Surface Plasmon resonance

• Surface plasmon resonance is the surface sensitive spectroscopic

method which measures the changes in the refractive index of the
medium directly in contact with nanoparticle surface.
• The localized confined to surface and interact with light resulting in
polarities.
• Propagating electron density waves occurring at the interface
between metal and dielectric.
The Kretschmann setup
Detector

• A nanoparticle film are deposited on the base of a dielectric prism.

• A beam of polarized light is transmitted through the prism.
• At a specific angle greater than the angle of total internal
reflection, a sharp minimum is observed in the reflection
coefficient, due to losses in the metal.
• The surface plasmons are electromagnetic waves that propagate
parallel to the metal surface from that interface the plasmons are
created.
SPR sensitive materials
• Gold (Au)
• Silver (Ag)
• Palladium (Pd)
• Tin (Sn)
• Copper (Cu)

SPR application
SPR is sensitive to its local environment such as refractive index of the
surrounding medium, temperature, pressure, humidity, pH etc
• pH sensor
• Temperature sensor
• Pressure sensor
• Protein detection

Advantage of Surface Plasmon Resonance technique

• Label free detection

• Reusable sensor
• Real time monitoring

Limitation of Surface Plasmon Resonance technique

• Very limited to choice of metals such as gold and silver.

• Sample need to be homogenous
• Refractive index is temperature dependant
